Authorities are urging people to avoid the West Side Highway area while they deal with the incident.
One person has been taken into custody, police have confirmed.
US media is reporting five people have been shot and that the incident happened near Stuyvesant High School, a top-ranked public New York City school.
The New York Fire Department said officials responded shortly after 3pm to reports of a mass casualty at Chambers and West Streets, near where the school is located.
The police say they are not looking for additional suspects.
