Lamela was set to start on the bench for Tottenham’s trip to Old Trafford.
But the London club have confirmed his absence just minutes before kick off.
“A late change to our bench as Erik Lamela is injured during the warm-up and is replaced by Luke Amos,” a statement on Twitter read.
Lamela, 26, has made just one appearance for Spurs so far this season.
He came on with 17 minutes remaining of their 3-1 home win over Fulham last week.
The Argentine was not included in the squad for Spurs’ opening-day win at Newcastle a fortnight back.
Tottenham did not sign a single player in the summer as manager Mauricio Pochettino was forced to contend with what he had.
The north London side will compete in the Champions League this season and are expected to challenge for the title.
Lamela played 33 games for Spurs last season as they secured second spot in the top flight.
As for Amos, he came on as a substitute in the win over Newcastle – albeit in the 88th minute.
The 21-year-old spent the second half of last season on loan at Stevenage where he helped the club avoid relegation to the Conference.
