The veteran cornerback suffered a hamstring strain in practice on Friday and “progress back next week,” according to the team.
The injury is expected to sideline Sherman for at least a week, which means the 30-year-old will sit out the clash against the Dallas Cowboys on Thursday.
Sherman felt his hamstring tighten up after falling during Friday’s practice and was forced to leave the field.
He has only recently joined the 49ers on a three-year deal, reported to be worth just over £30million after being released from NFC West rivals the Seattle Seahawks.
Sherman was recovering from surgery to repair a ruptured achilles but was cleared by 49ers medical staff a day before the team’s training camp started.

49ers coach Kyle Shanahan recently admitted that he wasn’t initially a “huge fan” of Sherman but has since he has changed his view after he joined the team.
“I’ve played against him a ton and wasn’t a huge fan of him. Always respected him, but he drove me crazy just like he’s probably driven every Niners fan crazy,” Shanahan told KNBR.
“You get an impression of guys when you see them on TV and stuff.
“I know he’s been outspoken and things like that so I know everyone has their own opinion of that, but you don’t know someone until you talk to them.”
Sherman has recently criticised the NFL over its new helmet-hit rule, which he labelled “ridiculous” and said the implementation of the regulation is “going to be a disaster”.
“They’ll see how ridiculous it is, once they make the refs call it,” Sherman said.
“It’s going to be worse than holding, worse than the catch rule.
“On a good form tackle, guys will lead with their shoulder pads, but you bring your head.
“It’s going to be a disaster.”
